    Isolation and Identification of fungi Associated with the Rhizosphere and Rhizoplane of Tobacco (Nicotiana tabacum)

    Fungi present in the rhizosphere and rhizoplane soils of tobacco (Nicotiana tabacum) were isolated and identified usin g standard methods. The fungi were Aspergillus niger, A. terreus, A. oryzae and Schizo saccharomyces Pombe. However, A. oryzae was absent from the rhizoplane soil but present in the rhizosphere soil has average pH value of 5.8, while that of the rhizoplane soil was 6.2. The average percentage percentage moisture content of the rhizophere soil was 10.27% and higher than that of the rhizoplane soil (8.80%). The fungi isolated are not harmful or pathogenic to tobacco.

    Isolation and Molecular Identification of Bacteria Associated With Soil Surrounding The Root of Citrus (Citrus sinensis L.) Tree

    Microbiological processes occur in the soil around the roots of land plants. This dynamic area, where plants and microorganisms interact, is crucial for the health and productivity of these plants. This study was aimed at isolating and molecularly identifying bacteria associated with the soil surrounding the root of an orange tree growing in the University of Ilorin, Nigeria to be able to know their roles, whether beneficial or harmful. Securing the soil is a way of having a food-secured world. Soil samples were collected at four different points from an orange tree trunk into separately labeled Ziploc bags using a separate sterile hand trowel for each collection. The labeled ziploc bags were brought to the University of Ilorin's Biology Laboratory for physicochemical analysis of the soil sample and isolation of bacteria using serial dilution method. Results of the physicochemical assessment of the bulked sample indicated that it is a sandy-loam soil with pH value of 6.7 (slightly neutral). Four bacterial species, i.e. Bacillus cereus, Lysinibacillusmacroides, Leucobacterkomogatae, and Alcaligenesfaecalis were isolated and identified molecularly. According to existing literature, the four bacterial species identified in this study are known for their abilities to enhance plant development and solubilize phosphorus, which are crucial for improving supply of nutrient.

    Microorganisms associated with some selected leafy vegetables sold in some markets in Ilorin, Kwara State

    Microorganisms associated with some commonly consumed leafy vegetables in popular markets of Ilorin, kwara State were investigated. The leafy vegetables were Amaranthus hybridus, Brassica oleraceae and Telfairia occidentalis. Seven species of bacteria were isolated namely Escherichia Coli, Staphylococcus aureus, Bacillus subtilis, Pseudomonas aeruginosa, Enterobacter aerogenes, streptococcus faecalis and Micrococcus luteus. Six species of fungi were equally isolated from some of the same leafy vegetables namely Aspergillus niger, A. Flavus, Rhizopus stolonifer, Neurospora sitophila, penicillium chrysogenum and Mucor racemosus. The isolates which are pathogenic might have arisen from conditions in the field and gardens where the leafy vegetables were grown, during transportation of the leafy vegetables to the market or as a result of the filthy environment in the markets where the leafy vegetables were obtained for this study. Therefore there is a great need for increased awareness on environmental hygiene in those fields and gardens and the market as well as sensitization of the farmers and traders on the need to adopt the modern technologies of transporting, storing and handling leafy vegetables.

    Molecular Characterization of Potential Crop Pathogens Associated with Weeds as Endophytes in Uniilorin Plantations, Nigeria

    Crop diseases are usually caused by inoculum of pathogens which might exist on alternate hosts or weeds as endophytes. These endophytes, cum pathogens, usually confer some beneficial attributes to these weeds or alternate hosts from protection against herbivores, disease resistance, stress tolerance to secondary metabolites production. This study was therefore carried out to isolate potential crop pathogens which exist as endophytes on weed species in the University of Ilorin plantations. Green asymptomatic leaves were collected from 10 weed species across the plantations, and processed for their endophytic fungi isolation. Isolates were purified into pure cultures and used for molecular identification using the internal transcribed spacer (ITS) region of the ribosomal DNA. Phylogenetic analysis of the fungal sequences using MEGA software revealed 9 fungal genera belonging to 13 species, with species in the genera Curvularia, Epicoccumand Daldinia occurring in more than one weed species, while other genera such as Alternaria, Fusarium, Chaetomium, Macrophomina, Arthrinium and Phomopsis occurred in just one weed species each. Daldinia eschscholtzii was isolated in this study as an endophyte from Loudetia arundinacea for the first time. This plant is very abundant in Nigeria and Africa where it is used majorly for thatching and feeding livestocks. This also represents the first endophytic fungi from the genus Loudetia. Potential relationship between the occurrences of these fungi as endophytes and as pathogens are discussed. These discoveries represent the first large-scale molecular identification and several first reports of endophytes from these weed species. These results also represent the first records of some of these fungi in Nigeria.

    Rhizosphere and non - rhizosphere soil mycoflora of corchorus olitorus

    The physicochemical and microbial analyses of the rhizosphere and non-rhizosphere soils of Corchorus olitorius (Jute) were conducted. The soil samples were analyzed before planting of Jute seeds and the average values of the parameters were 11.24% (percentage moisture content), 0.29ml/g (water holding capacity), 1.36% (organic matter content) and 6.80 (soil pH). The textural class of the soil sample is sandy. Aspergillus niger and Rhizopus stolonifer occurred most followed by Aspergillus flavus, while Penicillium chrysogenum, Saccharomyces cerevisiae and Neurospora crazza occurred least in the rhizosphere and non – rhizosphere soil samples of Jute in this study. Occurrence of these fungi in both the rhizosphere and non-rhizosphere soils varied widely throughout the duration of this study. It can be concluded that the jute plants benefitted from the microorganisms (fungi) in their rhizosphere and non – rhizosphere zones since the plants showed healthy growth.
