A.; Nuhu, K.M.; Babatunde, A.Item Comparative Antileukemic Activity of a Tetranorditerpene Isolated from Polyalthia longifolia Leaves and the Derivative against Human Leukemia HL-60 cells(Japan Oil Chemists’ Society., 2017) Afolabi, S.; Olorundare, O.; Ninomiya, M.; Babatunde, A.; Mukhtar, H.; Koketsu, M.Abstract: The discovery of potent cytotoxic isolates from botanicals provides an opportunity to explore this viable tool for cancer chemoprevention. The antileukemic potential of clerodane diterpene from Polyalthia longifolia leaves has already been established. However, in this present study, utilizing chromatographic techniques we report for the first time, the isolation of a rare tetranorditerpene (compound 1) from P. longifolia. The structure of compound 1 was elucidated and confirmed by spectrophotometric data. UPLCMS analysis was conducted on the methanolic extract, ethyl acetate fraction, and isolated tetranorditerpene showed that the tetranorditerpene is one of the major constituents of the plant with a retention time of 30.78 min. In addition, a methyl ester derivative (compound 2) of the isolated tetranorditerpene was synthesized. Using the CCK-8 assay, we compared the cytotoxic potential of isolated tetranorditerpene (1) and methyl ester derivative (2) with the previously isolated clerodane diterpenes. Our results showed that the methyl ester derivative (2) displayed the highest inhibitory activity against human leukemia HL-60 cells. The isolated tetranorditerpene (1) did not exhibit significant inhibitory effect against HL-60 cells. Morphological examination indicated chromatin condensation and nuclear fragmentation suggesting induction of apoptosis in compound 2 treated HL-60 cells. The methyl esterification of the isolated tetranorditerpene (1) conferred on it a significant level of antileukemic activity suggesting the possibility of a synergistic relationship between pure compound isolation and synthetic reaction in the discovery of new chemopreventive agents.Item Fibrinolytic proteins of normal pregnancy and pre-eclamptic patients in north west Nigeria(Makerere University, 2018) Oladosu-Olayiwola, O.; Olawumi, H.; Babatunde, A.; Ijaiya, M.; Durotoye, I.; Biliaminu, S.; Ibraheem, R.Abstract Background: The hypercoagulability of pregnancy is exaggerated in pre-eclamptic state because of endothelial activation with resultant production of some endothelial derived proteins that are said to be inhibitors of fibrinolysis. This study compares these proteins like tPA, PAI-1 and D-dimers in normal pregnant women and the pre-eclamptic women. Methodology: This was a comparative cross-sectional study. Eighty-five pre-eclamptic women were recruited as subjects and eighty five age, trimester and parity matched normotensive pregnant women as controls. Levels of PT, aPTT, tPA, PAI-1, D-dimer protein were determined in blood samples of subjects and controls. Urinalysis was performed with dipstick method on their urine samples. Data generated was analysed using the IBM®SPSS 20.0 (2011) soft ware packages and the level of significance was a p-value <0.05. Results: The mean age of the respondents was 29.9±5.2 years. The median(25th-75th percentile) values of D-dimer, tPA, and PAI-1 of subjects were 730 (305.000-1560.000ng/ml), 0.11 (0.065-0,300ng/ml) and 3.65 (2.970-4,400ng/ml) respectively which were significantly higher than the corresponding values in the controls of 520 (24.000-1030.000ng/ml), 0.05 (0.040-0.090ng/ml and 2.650 (2.125-3.400ng/ml) respectively, p<0.05 each. Conclusion: The abnormal levels of PAI-1,D-dimer and tPA imply that they contribute to the exaggerated hypercoagulabilty state in pre-eclampsia thus, measuring their levels can help in the management of the condition.Item Pattern of haematological malignancies in Ilorin, Nigeria: a ten year review(Internet Scientific Publications, 2008) Babatunde, A.; Amiwero, C.; Olatunji, P; Durotoye, I.A 10 year retrospective survey of haematological malignancies in Ilorin, Nigeria, was carried out based on data from the Records Department and the Cancer Registry of the Morbid Anatomy Department of the University of Ilorin Teaching Hospital.The aim was to determine the current pattern of haematological malignancies in the study area and compare with previous reports from other parts of Nigeria and worldwide.A total of 370 cases of haematological malignancies which were diagnosed within the study period (January 1996 to December 2005) by means of available haematological laboratory methods and histological examinations were analyzed. The distribution of the various haematological malignancies recorded were: ALL 18(4.9%), AML 18(4.9%), CLL 20(5.4%), CML 42(11.4%), Hairy cell leukaemia 2(0.5%), NHL 104(28.1%), Hodgkin’s disease 42(11.4%), Burkitt’s lymphoma 102(27.5%), Multiple myeloma 20 (5.4%) and Plasma cell leukaemia 2(0.5%)The lymphomas were found to constitute the highest prevalence (67.0%), with non-Hodgkin’s lymphoma being the commonest, accounting for about 42% of the lymphoma cases. Hairy cell leukaemia and plasma cell leukaemia were the least seen, each constituting 0.5% of the cases.The distribution of the various haematological malignancies in this study is similar to the pattern reported in previous studies, although, there appears to be generally a lower prevalence of haematological malignancies recorded when compared to the earlier findings.Difficulties in management and poor outcome observed for the various haematological malignancies are attributable to late presentation, high patients default rate, poverty and shortage of chemotherapeutic agents.Item Polyalthia longifolia extract triggers ER stress in prostate cancer cells concomitant with induction of apoptosis: insights from in vitro and in vivo studies(Hindawi publishers, 2019) Afolabi, Saheed O.; Olorundare, Olufunke E.; Babatunde, A.; Albrecht, R.M.; Koketsu, M.; Syed, Deeba N.; Mukhtar, HassanPlant-based therapies are being explored to prevent or treat several cancer types. The antioxidant properties of Polyalthia longifolia plant are well established. In our previous work, we demonstrated the presence of cytotoxic compounds in the methanol extract of Polyalthia longifolia (MEP) with potent activity against human leukemia cells. In the present study, we evaluated the efficacy of MEP against prostate cancer (PCa) and established the molecular basis of its effect in in vitro and in vivo models. We observed that MEP treatment resulted in a significant decrease in the growth and viability of PCa cells, associated with arrest in the G1/S phase of the cell cycle. Apoptosis was confirmed as the primary mode of MEP-induced cell death through activation of the intrinsic apoptotic machinery. Proteomic and biochemical studies identified BiP as an important target of MEP with the activation of the ER stress pathway, as a potential mechanism driving MEP-induced apoptosis. The extract exhibited strong efficacy in the PCa xenograft mouse model with significant inhibition of tumor growth and reduced tumor burden. Taken together, our findings indicate that MEP-induced apoptosis in PCa cells concomitant with the activation of the ER stress pathways results in the inhibition of tumor growth, in vitro and in vivo. Our studies provide initial evidence of the efficacy of MEP against PCa and advocate for in-depth studies in other preclinical models for its possible use in clinical settings.Item Unusual Presentations of Multiple Myeloma: Case Report And Review With Focus On Therapeutic And Psychological Aspects(Internet Scientific Publications, 2006) Agaja, S.; Babalola, A.; Olawumi, H.; Babatunde, A.Background: Unusual presentations of multiple myeloma which poses a diagnostic dilemma. Objective: To report unusual presentations of multiple myeloma and highlight how early detection of these unusual features will encourage early investigation, diagnosis and appropriate management and consequently better prognosis of this difficult disease of multiple myeloma. Patients and Methods: Two cases of multiple myeloma initially managed at Ela Memorial Medical Centre, Ilorin, were presented. The clinical features, management and outcome were extracted from the case notes. After operations on the patients the histological specimens were sent to University of Ilorin Teaching Hospital (U.I.T.H.). The patients were later transferred to UITH for further management. Results: The first case report presented with multiple swellings on the right shoulder, left chest wall, and back and weight loss. X-ray showed fracture of the left clavicle. The erythrocyte sedimentation rate was normal. Bence-Jones Protein was positive and histological report and Bone Marrow aspiration cytology confirmed multiple myeloma. The second case report presented with inability to use the right upper limb and later developed severe pain on the right shoulder and weakness of the right upper limb. X-ray showed fracture of the upper third of the right humerus. Erythrocyte sedimentation rate was very high and Bence-Jones protein was negative. Histological report was suggestive of multiple myeloma whilst bone marrow aspiration confirmed multiple myeloma. The first case was lost to follow up whilst the second case died during chemotherapy. Conclusion: Presence of multiple swellings on the body and pathological fractures should suggest that such patients should be investigated for multiple myeloma. Failure to start the investigations will lead to delayed diagnosis, delayed management and very poor prognosis.
