    Cyanide-induced hyperthyroidism in male Wistar rats.

    Background: Cyanide is one of the major environmental pollutants termed thyroid disruptor. Regardless of its origin, it is a primary toxic agent. This study was designed to understand the impact of prolonged low dose cyanide exposure on the structure and function of the thyroid gland. Materials and Methods: Twelve F1 male Wistar rats were used for this study. They were divided into two groups of six animals each. The first group served as the control group and received 0.25M sucrose while the second group being the treated group received 2 mg/kg body weight (BW) potassium hexacyanoferrate III solution. The treatment duration was 56 days following which the animals were sacrificed by cervical dislocation. Blood samples were drawn to determine serum FT3, FT4 and thyroid stimulating hormone (TSH) levels. The thyroid gland was also excised and processed for light microscopic studies. Result: An increase in serum FT3 and FT4 with decrease serum TSH was obtained in the treated group. Application of one-way analysis of variance (ANOVA) statistical analysis showed that there were highly significant differences ( P < 0.05) in the activities of FT3, FT4 and TSH when compared with those of the control group. Light microscopic examination of thyroid gland from the treated group revealed marked epithelial hyperplasia with cellular degeneration and scanty cytoplasm while the control group revealed normal thyroid architecture. Conclusion: Results obtained revealed that hyperthyroidism was induced by cyanide. Results obtained revealed that hyperthyroidism was induced by cyanide.

    Histological study of smoke extracts of Tobacco nicotiana on the heart, liver, lungs, kidney, and testes of male Sprague Dawley rats.

    Background: Some of the effects of tobacco on man’s health are well documented in many scientific reports. Whenever tobacco is used either in smoked or chewed form, nicotine is absorbed by the lungs and oral cavity and is spontaneously moved into the bloodstream where it is circulated throughout the body system. Materials and Methods: Ten male Sprague-Dawley rats were used for this investigation. The animals were randomly assigned into two groups, A and B, of five animals each. The animals in group B (treatment group) were exposed to smoke from a completely burnt 0.74 g leaf extract of Tobacco nicotiana, wrapped in 0.5 g of sterilized cotton wool for 5 minutes three times daily (7 am, 10 am, and 1 pm). The animals in group A (control group) were exposed to smoke from completely burnt 1.24 g of sterilized cotton wool with the same parameters as observed with the treatment groups. The duration of exposure was 5 days. Three hours after the last exposure, all the animals were killed by cervical dislocation. The heart, liver, lungs, kidney, and testes were carefully excised, blotted dry, and fixed in formol saline for histological analysis using Hematoxylin and Eosin stain. Results: Using the light microscope, it was observed that the histoarchitectural profiles of the studied organs in the sections obtained from the control animals were well preserved. Histopathological observations of the heart, liver, lungs, kidney, and testes in the treated animals showed a varying pattern of histological alterations, and distortions such as mild edema and occasional destruction of myocardial fibers, degeneration of the hepatocytes, reduction in the population of the germ cells, enlargement of the alveoli, alveolar hemorrhage, shrinkage of the glomerulus and glomerular hemorrhage were observed in the sections of the organs of the study of the animals in the treatment group when compared with the control group, hence showing that the smoke extract of Tobacco nicotiana has adverse and compromising effects on the heart, liver, lungs, kidney, and testes of male Sprague-Dawley rats. Conclusion: From these observations, it can be inferred that the exposure of male Sprague-Dawley rats to the smoke extract of Tobacco nicotiana may be associated with structural damage of some vital organs.

    Some of the effects of Aqueous Leaf Extract of Cnidoscolous aconitifolius (Euphorbiaceae) on the Morphology and Histology of the kidney, and Liver of Sprague Dawley Rat.

    Cnidoscolus aconitifolius belongs to a group of arbrescent shrubs. It has continued to be used as food, medicine and ornamental plant till date.Awide variety of claims have been made for its medicinal efficacy as a treatment for numerous ailments ranging from its ability to strengthen fingernails and darken gray hair to curing alcoholism, insomnia, gout, and scorpion stings. This study was to elucidate the effect of aqueous leaf extract of Cnidoscolus aconitifolius commonly used as food supplement on the kidney and liver of Sprague Dawley rats. Thirty-two Sprague Dawley rats of both sexes (with an average weight of 235g) were randomly assigned into one control group (n=8) designated as A, while the extract treated groups consists of 3 subgroups and were designated as B, C, and D. the animals in the extract treated groups B, C and D were administered 600mg/kg, 400mg/kg and 200mg/kg bwt of the extract respectively, while the control group received equal volume of phosphate buffered saline (PBS). The administration was done orally using an orogastric tube for seven days (7d). Twenty-four hours after the last administration, all the animals were sacrificed by cervical dislocation. Laparatomy was performed and the liver and kidney excised, trimmed free of fat, rinsed in cold phosphate buffered saline solution. The liver was quickly fixed in 10% formolsaline, while the kidney was fixed in Bouin's fluid for histological processing. The histological findings after Hematoxylin and Eosin (H&E) staining showed that the extract treated sections the Kidney and Liver has no compromising effect on the cellular integrity of the studied organs. There were no degenerative changes and no intercellular vacuolations in the renal cortex and liver parenchyma. These findings indicate that the administration of aqueous leaf extract of Cnidoscolus aconitifolius has no adverse effects on hepatic and renal morphology of the treated Sprague Dawley rats. It is therefore recommended that further studies at corroborating these observations be carried out.
